Zynga Dealing With Insider Trading Charges - Business Insider

Zynga's stock has crated to around $3 from an IPO price of $10.

Or, more importantly, it's now under $3, down from $12 when Zynga had a secondary offering of shares in April earlier this year.

During that secondary offering, insiders like Mark Pincus and a number of investors cashed out, selling about $516 million in stock.

Then Zynga had a disastrous second quarter where it reduced its full-year guidance, sending the stock tumbling.

Because the timing of the secondary offering, much of the Internet ? which already loves to hate on Zynga ? is starting to wonder if Zynga's top executives knew something the rest of the world didn't know.

And now, as to be expected in a situation like this, a law firm has filed an insider trading lawsuit against Zynga, Ben Popper of The Verge reports.

Anything's possible, but we highly doubt Zynga's top executives would really do something as colossally stupid as commit insider trading.

First, for these guys, it's not a lot of money in the big picture. Much of their wealth is still locked up in Zynga's stock.

Second,?if Zynga's insiders had some kind of crystal ball telling them that Facebook games were going to decline as much as they did, dealing a crippling blow to their business,?they would have put much, much more emphasis on mobile.

Zynga is?now?focusing more on its mobile games and it is planning on releasing real-money gaming properties, a nod at a billion-dollar opportunity in online gambling.?Pincus has said Zynga is working on several iterations of the Draw Something franchise and the With Friends franchise, its most popular mobile franchises.?

Pincus also said Zynga sees "opportunities to grow bookings to move male-oriented games," referring to hardcore games like those produced by Kixeye. This is indicative of a change in strategy when it comes to Facebook, but it's only coming?after?its weak second quarter.

None of this reads like an executive team that is ready to abandon the company after cashing out a little bit of stock.

People make terrible mistakes all the time, even for relatively small sums of money. No doubt, these guys could have screwed up to quickly pocket millions.

Unless Pincus turns out to be some kind of James Bond-level villain (which remains to be seen), we think he and his team are more interested in building a lasting business than cashing out short-term for a few million dollars.?

Uganda says avoid hand shakes as Ebola returns

KAMPALA (Reuters) - Ugandan President Yoweri Museveni on Monday advised people to avoid shaking hands, casual sex and do-it-yourself burials to reduce the chance of contracting the deadly Ebola virus after an outbreak killed 14 people and put many more at risk.

Museveni's advice came as scared patients and health workers fled a district hospital in rural western Uganda where several cases of Ebola were being treated and as the authorities tried to alter people's behaviour to stop the virus spreading.

"We discourage the shaking of hands because that can cause contact through sweat which can cause problems ... and avoid promiscuity because these sicknesses can also be transmitted through sex," Museveni said in a public statement.

There is no treatment for Ebola, which is transmitted by close contact and body fluids such as saliva, vomit, faeces, sweat, semen and blood. The authorities fear a repeat of an outbreak in 2000, the most devastating to date, when 425 people were infected, more than half of whom died.

Health workers suspected that the latest outbreak - which was confirmed on Friday - had occurred about three weeks ago in Nyanswiga village, Museveni said, adding that doctors had initially thought the symptoms were atypical of Ebola.

Nyanswiga, in Kibaale district, is about 170 km (100 miles) west of the capital Kampala, near the Democratic Republic of Congo where the virus first emerged in 1976, taking its name from the Ebola River.

The World Health Organisation has said the origin of the latest outbreak has not yet been confirmed, but that 18 of the 21 confirmed cases so far were understood to be linked to one family.

One of the 14 people reported to have died was a local health worker, Clare Muhumuza. She was transferred to Mulago hospital in Kampala where she died, stoking fears the disease could spread in the Ugandan capital.

"EVIL SPIRITS"

Museveni urged people to also be cautious when it came to burying people, saying that people had contracted the virus while burying Ebola victims because they remained contagious even when dead.

"In case somebody dies from what you suspect to be Ebola please do not take on the job of burying him or her, call the medical workers to be the ones to do it because they are the ones that can do it safely," he said.

Christine Ondoa, Uganda's minister for health, told a news conference in Kampala that people had initially put off seeking medical care because they believed the virus was the work of "evil spirits".

According to Tumusiime Jamilo, a reporter with Kagadi Kibaale Community Radio (KKCR), panic had gripped the Kagadi hospital in Kibaale, where suspected Ebola cases were being treated.

Some hospital staff had initially fled the establishment but were now returning as the authorities were providing protective gear for them, he told Reuters.

Kibaale local government authorities have ordered the closure of local primary and secondary schools and banned public gatherings as a precaution, he added.

Dan Kyamanywa, a health officer in Kibaale district, said up to 80,000 people in the district were at risk of infection.

Ebola symptoms include the sudden onset of fever, intense weakness, muscle pain, a headache and a sore throat, followed by vomiting, diarrhoea, rashes, impaired kidney and liver functioning and both internal and external bleeding.

Depending on the strain, the virus kills up to 90 percent of those who contract it.

Australia's major sporting event industry will be showcased this weekend at a high-level networking reception in London as the Olympic Games gets underway.

Hundreds of dignitaries and decision-makers from future host countries will attend Australia Unlimited 2012, a multi-media event featuring interactive and audio-visual displays at Australia House, the Australian High Commission, on Saturday night London time.

Minister for Sport, Kate Lundy, will deliver the keynote address profiling the achievements of Australia's world-leading major sporting event companies, and launch an innovative app detailing Australian companies with experience in the sporting event industry.

Trade and Competitiveness Minister Craig Emerson said Australia's expertise in sports event organising was further proof of the diversity of its economy and of its exports base.

"This industry is important not only for the export revenue and jobs it creates, but also for its role in promoting Australian professionalism and skill in the biggest arena of all?international sport," he said.

"Following our successful staging of the Sydney 2000 Olympics and subsequent sporting events, Australia has established a distinguished track record in helping other host nations achieve similar results," Dr?Emerson said.

The guest list for Australia Unlimited 2012 includes delegates from countries that are either hosting or bidding to host forthcoming Olympics, FIFA World Cups, Rugby World Cups and other major sporting events.

Among them will be representatives from Brazil, Japan, Qatar, Russia, Spain, South Korea, Turkey and the United Kingdom.

Minister for Sport Kate Lundy said the reception was a chance to mark Australia's contribution to the London Games through projects such as the London Olympic Stadium, designed by Populous, the Olympic Village, built by Lend Lease, and the blue hockey pitches, produced by Melbourne-based manufacturer Advanced Polymer Technology Australasia.

"This event is a wonderful initiative designed to highlight the strength of Australia's sport industry," Senator Lundy said.

"In Australia we have a breadth of expertise that has fostered our international reputation as the go-to nation for hosting world-class sporting events.

"Our major sporting event capability covers the full 10-year event lifecycle, including bidding, planning, staging, management and venue design and construction."

Minister Lundy will launch an innovative technology app called Track Record at the event. The app profiles 76 Australian firms with an unparalleled understanding and expertise in planning and staging major events.

The new app is compatible with both Apple and Android technology and available for download through iTunes and Google Play. In addition to the Track Record app, a special edition of the Australia Unlimited iPad magazine app will be showcased at the reception.

JACKSON, N.J. (AP) ? A boy is recovering after he was hit in the face by a bird while riding a roller coaster at an amusement park in New Jersey.

The collision occurred as the Kingda Ka coaster at Six Flags Great Adventure in Jackson was returning to the station Thursday.

Google acquires Mac, iOS email client Sparrow

Software developer Sparrow, maker of the popular Mac and iOS email clients of the same name, announced on Friday that it has been acquired by Google, and that the Sparrow team will be joining forces with the search giant.

A Google spokesperson said:?"The Sparrow team has always put their users first by focusing on building a seamlessly simple and intuitive interface for their email client. We look forward to bringing them aboard the Gmail team, where they'll be working on new projects."


Sources told?Computerworld UK's sister publication?Macworld?that while Google will continue to support Sparrow's apps, and provide updates related to security and bug fixes, there are no plans to provide significant updates to those apps. Sparrow had reported a couple months back that it was hard at work on an iPad version of the app, which has yet to be released.

On Sparrow's blog, CEO Dom Leca writes that Sparrow is "joining the Gmail team to accomplish a bigger vision--one that we think we can better achieve with Google."

Though Google isn't yet saying which products the Sparrow team will work upon, one wonders whether perhaps the company hopes to use its freshly-acquired apps to improve the rather unloved Gmail app for iOS.

(AP) ? Ernie Els plucked the ball from the hole after one last birdie and heaved it into the grandstand. At the time, it looked like nothing more than a classy gesture by a former British Open champion ? not the next one.

The name on the claret jug was supposed to be Adam Scott, who had a four-shot lead with four holes to play.

But in a shocking turnaround Sunday, Els returned to the 18th green less than an hour later to claim the oldest trophy in golf. Scott joined a list of players who threw away a major.

That was not lost on Els, whose heart sank when he looked over at the 32-year-old Australian.

"Sorry," Els told him. "You're a great player, a great friend of mine. I feel very fortunate. You're going to win many of these."

Scott might not get another chance like this.

After hitting a 3-wood into a pot bunker on the final hole, Scott had one last chance when he stood over a 7-foot par putt to force a playoff. It stayed left of the cup, and Scott dropped into a crouch. Standing off to the side, his chin quivered as the magnitude of the meltdown hit him. Instead, he mouthed one word: "Wow."

Wow, indeed.

Even though Els had gone more than two years without winning, and had thrown away two tournaments in recent months with shaky putting, the Big Easy felt all along that something special was going to happen at this British Open.

And it did ? all because of a collapse by Scott that no one saw coming.

"I know I let a really great chance slip through my fingers today," Scott said.

On a wind-swept afternoon at Royal Lytham & St. Annes that blew away the hopes of Tiger Woods and a handful of others, Scott looked steady as ever by going eight straight holes without making bogey. And that's when it came undone.

"I had it in my hands with four to go," Scott said.

A bogey from the bunker on the 15th cut the lead to three. That was followed by a three-putt bogey on the 16th, where his 3-foot par putt spun in and out of the cup and made the gallery gasp. From the middle of the 17th fairway, he hit a 6-iron that turned left, ran down the slope and took one last bounce in shin-high grass.

"I thought, 'Hold on. We've got a problem here,' " said Graeme McDowell, playing with Scott in the final group.

By then, Els had posted a 2-under 68 with a 15-foot birdie putt on the final hole, a cheer that Scott recognized while playing the 17th. Scott failed to get up-and-down for par from the rough and suddenly was tied.

Els headed to the practice green, where it rarely works out for him. In perhaps the most crushing defeat in a career filled with them, Els was on the putting green at Augusta National in 2004 when Phil Mickelson made an 18-foot birdie putt to win the Masters.

"I just thought, 'I'll probably be disappointed again,' " Els said. "You're not really hoping the guy is going to make a mistake, but you're hoping you don't have to go a playoff, you can win outright. This one was different, because I feel for Adam."

Els, who started the final round six shots behind, wound up with his second British Open ? the other one was 10 years ago at Muirfield ? and fourth major championship at a stage in his career when it looked as if his best golf was behind him.

"Amazing," Els said. "I'm still numb. It still hasn't set in. It will probably take quite a few days because I haven't been in this position for 10 years, obviously. So it's just crazy, crazy, crazy getting here."

The celebration was muted, unlike his other three majors.

"First of all, I feel for Adam Scott. He's a great friend of mine," Els said. "Obviously, we both wanted to win very badly. But you know, that's the nature of the beast. That's why we're out here. You win. You lose. It was my time for some reason."

The wind finally arrived off the Irish Sea and ushered in pure chaos ? a mental blunder by Woods that led to triple bogey on the sixth hole, a lost ball by Brandt Snedeker that took him out of contention and a topped shot that made McDowell, a former U.S. Open champion, look like an amateur.

"I guess my disappointment kind of seems relatively stupid in relation to the guy ... I've just seen a guy lose The Open Championship," said McDowell, who played in the final group of a major for the second straight time.

Nothing was more stunning than what happened to Scott, who closed with a 75.

"I managed to hit a poor shot on each of the closing four holes," Scott said. "Look, I played so beautifully for most of the week. I shouldn't let this bring me down."

Even so, it added another chapter to Australian heartbreak, most of that belonging to his idol, Greg Norman.

Scott was the fourth Australian since the 2007 Masters to lead going into the final round of a major, yet the proud land Down Under remains without a major since Geoff Ogilvy won the U.S. Open at Winged Foot in 2006.

"Greg was my hero when I was a kid, and I thought he was a great role model, how he handled himself in victory and defeat," Scott said. "He set a good example for us. It's tough. I can't justify anything that I've done out there. I didn't finish the tournament well today.

"But next time ... I'm sure there will be a next time and I can do a better job of it."

Already in the World Golf Hall of Fame, the 42-year-old Els joined even more elite company. He became only the sixth player to win the U.S. Open and British Open twice. The others are Jack Nicklaus, Woods, Walter Hagen, Bobby Jones and Lee Trevino.

Woods came undone on the sixth hole when he tried to blast out of a bunker from a plugged lie, stayed in the bunker, and three-putted for triple bogey. Still with an outside chance after a birdie on the 12th, he stuck with his conservative plan of hitting iron off the tee and made three straight bogeys. He closed with a 73 to tie for third with Brandt Snedeker, who also had his share of problems for a 74.

Woods had his best finish in a major since he lost to Y.E. Yang in the 2009 PGA Championship, though he remains winless in the last 17.

"It's part of golf," said Woods, who moves to No. 2 in the world. "We all go through these phases. Some people, it lasts entire careers. Others are a little bit shorter. Even the greatest players to ever play have all gone through little stretches like this."

Els finished at 7-under 273. He failed to qualify for the Masters this year for the first time in nearly two decades, but that won't be a problem now. His win gives him a five-year exemption into the majors.

It was the most shocking collapse at the British Open since Jean Van de Velde took a triple bogey on the final hole at Carnoustie and lost in a playoff. But this was different. It wasn't a last-minute blowup, more of a slow bleed, similar to Jason Dufner losing a five-shot lead to Keegan Bradley in the PGA Championship last year, or Ed Sneed making bogey on the last three holes at the 1979 Masters.

There was just enough wind to make the 206 bunkers at Royal Lytham look a little bit bigger. And as the gusts increased, a calm week turned chaotic.

It started with Woods on the sixth hole, his first triple bogey at a major championship since he lost his ball on the opening hole at Royal St. George's in 2003.

"One yard," he said to his caddie, a measure of the miss. It plugged near the steep wall of a pot bunker.

Instead of chipping to the middle of the bunker, Woods tried to get out with a ferocious swing. The ball smacked into the wall, nearly hit him and wound up near the left wall. He sat on the grass, his left knee (which has gone through four surgeries) flexed underneath him, his right leg extended as he dipped his upper body toward the sand to make a swing. This one also hit the wall, and caromed around and out to the right. From there, he three-putted for a 7.

"The game plan was to fire it into the bank, have it ricochet to the right and then have an angle to come back at it," Woods said. "Unfortunately, it ricocheted to the left and almost hit me."

Just like that, he was seven shots behind. It was the second time this year that one of golf's biggest stars made triple bogey in the final round of a major while in contention. Phil Mickelson made his on the fourth hole at the Masters and never recovered.

Els made a bogey on the ninth to fall six shots behind. All that did was fire him up, and he came home in 32. His 68 is best measured in these terms ? of the last 12 players who teed off in the final round, no one else had better than a 72.

Yet there was one more collapse, in the final hour, and it was the one everyone will remember from this British Open. It's one Scott will somehow need to forget. As winner and runner-up met in a portable trailer before going out to the trophy presentation, Els told him: "Don't beat yourself up."

(AP) ? Shortly after Penn State tore down its famed statue of coach Joe Paterno, the NCAA announced Sunday it would impose "corrective and punitive" sanctions against the university in the wake of a devastating report that asserted top university officials buried child sex abuse allegations against a retired assistant coach more than a decade ago.

The NCAA, acting with rare speed, said it will spell out the penalties on Monday. The governing body did not disclose further details.

If precedent holds from recent cases, Penn State will face a loss of scholarships and a multi-year ban from bowl games ? and with it, the financial windfall and showcase that comes with postseason play.

Yet NCAA President Mark Emmert cautioned last week that he hasn't ruled out the possibility of shutting down the Penn State football program altogether, saying he had "never seen anything as egregious" as the Jerry Sandusky sex abuse scandal.

A harsh penalty would have repercussions well beyond football, whose large profits ? more than $50 million, according to the U.S. Department of Education ? subsidize dozens of other sports programs at the school. The potential for a historic NCAA penalty also worries a region whose economy is built at least partially on the strength and popularity of the football program.

"It's going to kill our town," said Derek Leonard, 31, a university construction project coordinator who grew up in the area.

Emmert has seemingly put the Penn State matter on the fast track. Other cases that were strictly about violating the NCAA rulebook have dragged on for months and even years.

As Penn State awaited its fate, construction workers took down the larger-than-life monument to its Hall of Fame coach ? on the six-month anniversary of his death from lung cancer at age 85.

The Paterno family released a statement criticizing Penn State's decision to remove the statue, saying it was made in haste and before all the facts about Paterno's role in the Sandusky scandal were known.

"Tearing down the statue of Joe Paterno does not serve the victims of Jerry Sandusky's horrible crimes or help heal the Penn State community. We believe the only way to help the victims is to uncover the full truth," said the family, which has vowed its own investigation following the release of an investigative report by former FBI Director Louis Freeh that found that Paterno and three other top Penn State administrators concealed sex abuse claims against Sandusky.

"Despite (Freeh's) obviously flawed and one-sided presentation, the university believes it must acquiesce and accept that Joe Paterno has been given a fair and complete hearing," the statement said.

The bronze statue, weighing more than 900 pounds, was erected in 2001 in honor of Paterno's record-setting 324th Division I coaching victory and his "contributions to the university." Students chanted, "We are Penn State" as it came down Sunday morning.

Penn State President Rodney Erickson said he decided the sculpture had to go because it "has become a source of division and an obstacle to healing."

In Washington, the White House said President Barack Obama believed "it was the right decision."

But the vast majority of fans gathering outside Beaver Stadium to watch the statue's removal disagreed. At least one woman wept, others expressed anger at the decision, and nearly all said they continued to support their beloved "JoePa."

"I think it was an act of cowardice on the part of the university," said Mary Trometter, of Williamsport, who wore a shirt bearing Paterno's image. She said she felt betrayed by university officials who pledged greater transparency but then failed to announce its decision on the statute until workers arrived shortly after dawn to begin tearing it down.

In NCAA terms, the July 12 release of the Freeh report may have hastened the process for the slow-moving governing body for college sports.

Recent major scandals ? such as improper payments to the family of Heisman Trophy winner Reggie Bush while he was at Southern California, and players at Ohio State trading memorabilia for cash and tattoos ? have resulted in bowl bans and the loss of scholarships.

Current NCAA rules limit the so-called "death penalty" to colleges already on probation that commit another major violation. That was the case when Southern Methodist had its program suspended in the mid-1980s, the last time the punishment was imposed on a major college football program.

NCAA leaders have indicated in recent months they are willing to return to harsher penalties for the worst offenses.

"This is completely different than an impermissible benefits scandal like (what) happened at SMU, or anything else we've dealt with. This is as systemic a cultural problem as it is a football problem. There have been people that said this wasn't a football scandal," Emmert told PBS recently. "It was that but much more. And we'll have to figure out exactly what the right penalties are. I don't know that past precedent makes particularly good sense in this case because it's really an unprecedented problem."

Another question is whether Penn State ? and, by extension, Paterno, major college football's winningest coach ? will have to vacate any victories. Paterno won 409 games for the school in his 46 seasons as head coach. USC lost a national title when it went on probation and Ohio State vacated the 2010 season, including its victory in the Sugar Bowl over Arkansas.

ACC Commissioner John Swofford said he doesn't know what the penalties will be, though many in college sports have given some thought to what they should be.

"I think a lot of us in this profession wrestle with that, to a degree, because the Penn State situation is unprecedented. I don't know of anything to compare it to," he said Sunday. "So, it's uncharted waters. A tragedy from every angle."

Kayla Weaver, a Penn State senior and member of the dance team called the Lionettes, said an NCAA death penalty would not only force the football players to transfer, but it would also force program changes for cheerleaders, dancers and band members and would hurt season ticket holders.

"It could ruin everything that we've built here," said Weaver, 21, from Franklin Lakes, N.J.

On Twitter, Akeel Lynch, a running back recruit who played high school football in western New York, wrote: "I still bleed blue and white," while quarterback Matt McGloin wrote, "The hotter the fire, the stronger the steel."

Tight end Garry Gilliam tweeted, "No matter what happens, I'm staying at Penn State."

Emails prove Arizona's SB 1070 immigration law racially motivated

The American Civil Liberties Union of Arizona has released thousands of e-mails proving that SB 1070, the state's anti-illegal immigration law, was racially motivated.
The e-mails, which have been posted in full online and excerpted in the Arizona Republic, were sent to and from former Arizona state Sen. Russell Pearce, SB 1070's Republican author. In them, Sen Pearce compares undocumented immigrants to lepers and Islamic jihadists, claims that the children of "illegal immigrants" will be criminals, accuses immigrants of turning America into a "fractured nightmare of social unrest and poverty," and asserts that corruption is "their only known way of life." The ACLU of Arizona obtained the emails through a public records request. Dozens of the emails have been included in a legal filing in which the ACLU is trying to prevent the most controversial parts of the law from going into effect. SB 1070, the so-called "show me your papers law," makes it a crime for foreigners in the state to be without immigration documents and forces police to determine individuals' immigration status during a "lawful stop, detention or arrest" or during "lawful contact" when there is reasonable suspicion that the individual is in the country illegally. The law also cracks down on anyone who harbors, hires or transports undocumented immigrants. It is the second-toughest anti-illegal immigration law in the nation, after Alabama's. Supporters of the law say it only enforces existing federal law and is necessary in the face of an uncontrollable flood of "illegals" crossing into the state from Mexico. Critics counter that SB 1070 encourages racial profiling; some even go so far as to call the law racist. Those who claim the law is rooted in racial prejudice got a huge boost from the emails released by the Arizona ACLU. Here is an alarming look inside the mind of Sen. Pearce, in his own words: * "Can we maintain our social fabric as a nation with Spanish fighting English for dominance?... It's like importing leper colonies and hope [sic] we don't catch leprosy. It's like importing thousands of Islamic jihadists and hope [sic] they adapt to the American Dream." * "[Undocumented immigrants] create enclaves of separate groups that shall balkanize our nation into fractured nightmares of social unrest and poverty." * "We are much like the Titanic as we inbreed millions of Mexico's poor, the world's poor, and we watch our country sink." * "9,000 people [are] killed every year by illegal aliens," and "illegal aliens in the United States have a crime rate that's two-and-a-half times that of non-illegal aliens... their children are going to make a huge additional crime problem in the US." * "Last week, Denver's illegal aliens sang our national anthem in Spanish and bastardized the words of OUR county's most sacred song." * "Corruption is the mechanism by which Mexico operates. Its people spawn more corruption wherever they go because it is their only known way of life." * "Tough, nasty illegals and their advocates grow in such numbers that law and order will not subdue them. They run us out of our cities and states. They conquer our language and our schools. They render havoc and chaos in our schools." * "Battles commence as Mexican nationalists struggle to infuse their men into American government and strengthen control over their strongholds. One look at Los Angeles with its Mexican-American mayor shows you Vincente [sic] Fox's General Varigossa [sic] commanding an American city." (A reference to former Mexican President Vicente Fox and Los Angeles Mayor Antonio Villaraigosa). SB 1070 opponents slammed Sen. Pearce in the wake of the e-mail release. "This just shows who Russell Pearce really is," Randy Parraz, who led a successful effort to recall Pearce from the state Senate, told the Arizona Republic. "You can call it hate speech. [SB 1070] was about using this type of fear and hate for political purposes." But a spokesman for Jan Brewer, Arizona's Republican governor, dismissed the e-mails as an "ACLU smoke screen." "The ACLU's tactic is a smoke screen," Matthew Benson told the Arizona Republic. "By focusing on an individual legislator's e-mails, they intend to divert focus from SB 1070's simple common sense language, language that the overwhelming majority of Arizonans and Americans support, and language the Supreme Court unanimously upheld." The released e-mails may provide the necessary ammunition to those who seek to persuade courts to overturn SB 1070. How hosts recognize bacteria

ScienceDaily (July 20, 2012) ? We are surrounded by bacteria, viruses, fungi and parasites. The fact that we nevertheless do not fall prey to infections is thanks to certain cellular sensor molecules such as toll-like receptors (TLR), which recognize the molecular structure of pathogens and intercede by ensuring an often completely unnoticeable elimination of the invaders. Their immune-activating abilities were only detected in 1998, a discovery which was awarded with the Nobel Prize. Now, an international research team led by Prof. Dr. Carsten Kirschning of the Institute of Medical Microbiology at the University hospital Essen and the University of Duisburg-Essen and PD Dr. Hubertus Hochrein is examining the complex recognition of bacteria.

Their findings have been published in the research journal Science.

Our innate immune system is clever. Certain structures, which are characteristic of many microorganisms are recognized via TLR which trigger the necessary inflammatory response. However, the situation can become dangerous: since this receptor system is so sensitive, the immune response to serious infections may be over-exaggerated and miss the mark. Blood poisoning and frequently septic shock is the result.

Thus, the experts decided to take a closer look at the host recognition of Staphylococcus and E.coli bacteria, often the main agents of blood poisoning. They established that in the cell, TLR13 recognises the segment of bacterial ribosomal ribonucleic acid to which specific antibiotics such as Erythromycin also bind should the segment not have been altered by mutation or other modifications. The concrete ribonucleic acid is referred as 23S rRNA. Animal and human ribosomes do not bind Erythromycin to their own 28S rRNA because its structure resembles that of the 23S rRNA of resistant bacteria.

The new findings are significant for the treatment of bacterial infections and gaining a greater understanding of antibiotic resistance. In addition, these findings may be of help in the therapy of immunological overreactions and lead to new vaccination strategies.

The study was conducted by an international research team led by Prof. Dr. Carsten Kirschning of the Institute of Medical Microbiology at the University hospital, University of Duisburg-Essen, PD Dr. Hubertus Hochrein of Bavarian Nordic in Martinsried, Prof. Dr. Stefan Bauer of the Institute of Immunology at the Philipps University of Marburg and Prof. Dr. Hermann Wagner of the Institute of Medical Microbiology, Immunology and Hygiene at the Technical University of Munich. Further collaborating scientists came from Munich, Switzerland and Japan.

A research study led by the Peninsula College of Medicine and Dentistry, University of Exeter, and Boston University School of Medicine, in collaboration with a global consortium, has identified genetic markers that influence a protein involved in regulating oestrogen and testosterone levels in the bloodstream.

The results, published online in PLoS Genetics, also reveal that some of the genetic markers for this protein are near genes related to liver function, metabolism and type 2 diabetes, demonstrating an important genetic connection between the metabolic and reproductive systems in men and women.

The study was carried out in collaboration with the Framingham Heart Study and investigators from 15 international epidemiologic studies participating in the Cohorts for Heart and Aging Research in Genetic Epidemiology (CHARGE) consortium.

Sex hormone binding globulin (SHBG) is the key protein that carries testosterone and oestrogen in the bloodstream in both men and women. As the main carrier of these sex hormones, SHBG helps to regulate their effects in different tissues and organs in the body. In addition to effects on reproduction in men and women through regulation of sex hormones, SHBG has been linked to many chronic diseases including type 2 diabetes and hormone-sensitive cancers such as breast and prostate.

Previous family studies have demonstrated that approximately 50 per cent of the variation in SHBG concentrations in the bloodstream is inherited from parents, suggesting that SHBG levels are under significant genetic control. However, little has been known about the specific genes that influence SHBG levels.

Investigators examined human genomes from 21,791 men and women to determine which genes influence SHBG levels and validated the results from this genome-wide association study (GWAS) in an additional 7,046 men and women. They identified 12 single-nucleotide polymorphisms (SNPs), or DNA sequence variations, associated with the concentration of SHBG circulating in the bloodstream. Although these genetic variants only explain a small fraction of the sex hormone variability seen between individuals, they could provide insight into the diseases connected to sex hormone regulation.

The results showed that the SNPs that influence SHBG levels are near genes related to liver function, fat and carbohydrate metabolism and type 2 diabetes. In addition, there were genes that had stronger effects in one sex compared to the other.

"These findings highlight the diverse range of biological processes that may be impacted by sex hormone regulation," said Dr. John Perry of the Peninsula College of Medicine and Dentistry, University of Exeter.
